Comparative evaluation of methods for isolating small extracellular vesicles derived from pancreatic cancer cells

Abstract Background Small extracellular vesicles (sEVs) are nanosized involved in cell-to-cell communication. sEVs have been widely studied for clinical applications such as early detection of diseases and therapeutics. Various methods isolation using, but different may result qualities impact downstream analysis applications. Here, we compared current performed a comparative from supernatant cultured pancreatic cancer cells. Methods Ultracentrifugation, ultrafiltration co-precipitation concentration were firstly evaluated yield, size, morphology protein level pellets. Then, isolate obtained by four purification methods: size exclusion chromatography, density gradient ultracentrifugation, immunoaffinity capturing, analysed compared. Results For the process, ultracentrifugation method high quality capturing purest with less contaminants, while ultracentrifugation-based smallest size. Proteomic revealed distinct contents purified methods. Conclusions isolating derived cell line, is recommended sEVs, be applied obtaining controlled suitable studies requiring purity loss subtypes without specific marker.
